Screening of hydrogen-producing aciduric Enterobacter aerogenes
strain using He-Ne laser irradiation

Abstract: Enterobacter aerogenes was irradiated by He-Ne laser to optimize the laser mutagenesis condition,and to test the heredity,aciduric and hydrogen-producing potential of the mutant. An aciduric mutant with steady hydrogen-producing capability,which was able to grow at acidic pH of 3.0,was obtained. Its capability of hydrogen production was measured in the batch culture experiment. The maximum hydrogen yield and hydrogen production rate was 1781 mL/L and 240 mL/(L•h),which were 48% and 32% higher than those of the parent strain,respectively. The results demonstrated that He-Ne laser mutagenesis could be used in the field of hydrogen-producing microorganism.

摘要: 运用He-Ne激光辐照产气肠杆菌(Enterobacter aerogenes),对激光诱变参数进行优化,考察了突变株的遗传稳定性、耐酸性和产氢特性。结果筛选到一株遗传性状稳定的高产氢突变株,具有良好的耐酸性,在pH值3.0时仍能生长。通过间歇实验,产氢量和产氢速率分别达到1781 mL/L和240 mL/(L•h),比原始菌分别提高了48%和32%。说明该He-Ne激光诱变育种技术可以在产氢微生物领域中应用。
